Schematic representation of Adhesion-to-suspension transition (AST) in vitro. Ectopic expression of the four hematopoietic factors IKZF1, NFE2, BTG2, IRF8 in adherent cell lines is sufficient to induce adhesion-to-suspension transition (AST), promoting anchorage-independent growth in vitro. AST induction leads to YAP/TEAD-dependent loss of cell-matrix interaction and confers resistance to anoikis achieved through HBA1/2-mediated suppression of lethal reactive oxygen species (ROS).
